Panel lines

Is there a primer in panel lines?  I have searched threads on them but still have questions.  How does one lay them out? Pencil then ink?  How do you deal with the inevitable screw ups?  Can the ink be removed and reapplied without affecting the dope finish?  I am probably guilding the lilly, but I'm building a Brodak profile P-51B and want to attempt panel lines. I am planning to do an Excaliber III scheme that's hanging in the NASM.

I also am trying to figure out whether to use paint masks or vinyl for lettering and canopy outlines. This is not planning on being a 20 point finish, but I try to push myself on each new build.
